Surface modification via strain-promoted click reaction facilitates targeted lentiviral transduction

Since the last decade, the Lentiviral Vector has emerged as a promising vector for gene delivery. This is coming from its high efficiency to transduce a large variety of cells in vitro as well as in vivo. Moreover, the Lentiviral Vector has a high degree of design flexibility allowing to choice the way of gene expression in time and space. One of the modifiable parameters is the envelope of the vector. According the type of envelope modification, it is possible to widen the type of cell target or to restrict it to specific cells.

In this study, the scientists design new envelopes which can interact with cancer specific ligands and which could be also useful for vector purification and immunomodulation. First, they demonstrated that these new types of envelope have no negative effects in term of production and gene delivery function. Then, they show that these types of pseudotype enhance the transduction efficiency of cancer cell lines such as LNCaP or U-87 MG.

This work is a good illustration of the possibility given by the flexibility of LV pseudotyping. Furthermore, there are other pseudotypes which have already been validated in other field of research. For example, the point was made that the Mokola envelope allows to transduce specifically astrocytes. In this regard, GEG Tech is the only company which provides this type of Lentiviral Vectors.
